Spin 106 early-mid log phase S2 cells at 500 xg for 5 min.
Decant sup.
Resuspend cells in 1ml media containing:
250 ml conditioned media
750 ml Schneider’s Serum Free media
Place cells in 12-well TC plate
Add 10 mg dsRNA to cells dropwise
Swirl to mix
Incubate for 1hr. @ 25°C
Add 1 ml Schneider’s + 20% FBS to cells
Seal plate with breathable film
Incubate 4 days @ 25°C
